Vitrification Media for PN stage to Blastocyst Freezing

 

Vitrification is an ultra-rapid freezing technique that is used to freeze PN stage embryos to Blastocysts. This frozen embryos and blastocysts can be used later on to achieve pregnancy after thawing.

 

The media contains Hydroxypropyl Cellulose (HPC) and Trehalose as an alternate for HAS and Sucrose respectively, which reduces cell membrane rupture due to osmotic imbalance.

FAQs

We do not inspire to use specific cryo-storage devise. Any storage device can be used.

In thawing the Warming Solution-1 should be incubated at 37 ᵒC for minimum of 1 hour and maximum overnight and in freezing, Equilibrium Solution and Vitrification Solution must be kept at 25-27 ᵒC for 30 minutes prior to use. If the medias are incubated in CO2 Incubator than restrict the exposure by keeping the media tightly capped.

Maximum 3 embryos/oocytes/blastocysts are advisable for freezing in single cryo-storage device.

If it is Day-2 or Day-3 embryos then can be transferred immediately and if blastocysts then transfer after 2 hours.

Ideally after 1 hour of denuding, oocytes can be vitrify.

2 hours after thawing of oocytes ICSI can be performed.

There are no much studies available for the same but as warming media contains sucrose/trehalose and does not contain any intracellular cryo-protectant, media from any brands can be used. Many scientists have experienced equal results using different brands for freezing and thawing of the embryos.
